RESUMO

Uninfected alligators (Alligator mississippiensis) exhibited high constitutive levels of hepatic gene expression related to immune function, whereas the highest-expressed hepatic genes of uninfected mice were related to metabolism. Intraperitoneal challenge of mice with bacterial lipopolysaccharide results in dramatic inflammatory effects including peritoneal ascites, febrile response, dramatic alterations in electrophoretic serum profile, and mortality. In contrast, coelomic injection of alligators with 200× the murine LD50 of intraperitoneal bacterial lipopolysaccharide resulted in no changes in serum protein profiles, behavioral effects, mortality, and no coelomic ascites. However, injection of juvenile alligators with live bacteria resulted in a titer-dependent decrease in metabolic rate, as measured by oxygen consumption. These results are the opposite of those observed for mammalian and avian species. The decreased oxygen consumption was not accompanied by changes in heart or respiration rate, indicating that this phenomenon was not due to bradycardia or bradypnea. Interestingly, challenge of alligators with bacteria resulted in the complete expulsion of digestive tract contents within four hours. We interpret these activities as temporary minimization of other biological systemic activities to redirect and devote energy to immune function. The reallocation of resources within an organism to fight infection without increases in metabolic rate has not been described in other animals.

To assess skin color change in alligators, we maintained animals in differently lighted environments and also measured skin colors in an ontogenetic series of wild animals. Juvenile alligators maintained in black enclosures exhibited a gradual lightening of skin color when shifted to white enclosures, and these observed changes were reversible. A histological examination of the skins of alligators maintained in dark tanks showed that the dermis exhibited a dense layer of pigmented cells, while samples from the same animals in light environments exhibited a more diffuse pigmented layer. As alligators grow, they exhibit an ontogenetic loss of stripes that may aid in crypsis and predation. Hatchlings have intense black and yellow vertical stripes that darken with age; adults are a more homogenous black/gray color. Since alligators live in temperate climates and adults have lower surface area/volume ratios, which can be detrimental for the absorption of radiant energy, the darker color of larger animals may also aid in thermoregulation. Alligators at the northern end of their range, with colder climates, exhibited darker skin tones, and the ontogenetic extinction of stripes occurred at a more accelerated rate compared to animals in southern, warmer regions, supporting the idea that latitude-dependent ontogenetic color shift has a role in thermoregulation.

BACKGROUND: While many nursing students work during clinical semesters, little is known about employment characteristics and relationships among employment, academic success and other variables. PURPOSE: To describe the demographic, educational, and health-related characteristics of clinical nursing students and the relationships among employment, semester grades and other characteristics. METHOD: Descriptive, correlational. RESULTS: Participants were BSN students (N = 1014) from four southern US universities who were 22.6 years old (SD = 4.6) and most likely to be Caucasian (N = 832, 82%) never married (N = 852, 84%) females (N = 886, 87%) with mean GPA of 2.97 (SD = 0.61). Most students (N = 670, 66%) reported semester employment averaging 16.7 (SD = 8.3) hours/week. Although no relationship was found between hours worked and semester GPA (r = -0.017, p = .588), race/ethnicity (F [2, 1003] = 19.87, p < .0001) and nighttime sleep hours (F [3, 997] = 7.841, p < .0001) had significant effects. Students working in healthcare had higher GPAs (M = 3.09, SD = 0.61, p < .0001) than non-healthcare workers. Students working daytime (M = 3.04, SD = 0.65, p = .031) or irregular shifts (M = 3.04, SD = 0.56, p = .036) had higher GPAs than students working evenings. CONCLUSION: While employment status did not influence GPA, race/ethnicity and amount of reported sleep did. Additional research is needed to provide evidence-based advisement recommendations for employed students.

Low unit response rates can increase bias and compromise study validity. Response rates have continued to fall over the past decade despite all efforts to increase participation. Many factors have been linked to reduced response, yet relatively few studies have employed multivariate approaches to identify characteristics that differentiate respondents from nonrespondents since it is hard to collect information on the latter. We aimed to assess factors contributing to enrollment of prostate cancer (PCa) patients. We combined data from the North Carolina-Louisiana (LA) PCa Project's LA cohort, with additional sources such as US census tract and LA tumor registry data. We included specific analyses focusing on blacks, a group often identified as hard to enroll in health-related research. The ability to study the effect of Hurricane Katrina, which occurred amidst enrollment, as a potential determinant of nonresponse makes our study unique. Older age (≥ 70) for blacks (OR 0.65) and study phase with respect to Hurricane Katrina for both races (OR 0.59 for blacks, OR 0.48 for whites) were significant predictors of participation with lower odds. Neighborhood poverty for whites (OR 1.53) also was a significant predictor of participation, but with higher odds. Among blacks, residence in Orleans parish was associated with lower odds of participation (OR 0.33) before Katrina. The opposite occurred in whites, with lower odds (OR 0.43) after Katrina. Our results overall underscore the importance of tailoring enrollment approaches to specific target population characteristics to confront the challenges posed by nonresponse. Our results also show that recruitment-related factors may change when outside forces bring major alterations to a population's environment and demographics.

Pathogenic microbes require Fe and Zn for growth and proliferation. Upon infection, microbes produce proteins, called sidephores, designed to strip serum divalent metals away from host proteins. Higher vertebrates respond to infection by increasing the expression of proteins that sequester serum iron away from bacteria. As a result, host plasma Fe levels decrease during the initial phases of infection. This study was conducted to determine if the American alligator, an ancient reptile, exhibits the same innate immune mechanism to protect against in vivo microbial proliferation. Intraperitoneal injection of juvenile captive alligators with bacterial lipopolysaccharide (LPS) resulted in a time-dependent decrease in plasma Fe, as determined by inductively coupled plasma emission spectroscopy. Plasma Fe levels decreased by 5.9, 10.6, and 18.6% relative to untreated control levels at 3, 6, and 12 hr post-injection, respectively, and remained decreased by 12.0% at 48 hr. Alligators injected with pyrogen-free saline did not exhibit statistically significant changes in plasma Fe concentrations at any time point observed. In contrast, serum Zn and Cu remained unchanged relative to untreated controls. To insure that the decreases in plasma Fe were not due to the repeated blood collections during the course of the kinetic study, another experiment was conducted in which plasma metals were measured at 24 hr post-injection. Once again, plasma Fe was reduced by 30.2%, whereas Zn and Cu did not exhibit appreciable changes. These results show that alligators exhibit low plasma Fe levels during an inflammatory response induced by bacterial lipopolysaccharide.
